Mitchell was born in Melbourne, Victoria, Australia and had a "hippie-ish" upbringing by her parents, who divorced during her childhood.[1] Her mother is a model turned designer and her father is a filmmaker.[2] She attended the St Michael's Grammar School. Her first name, "Radha", is of Indian Hindu origin, where the name is described to mean the paramour of Lord Krishna.[3] Mitchell is a vegetarian and practices Yoga.

Mitchell got her start in acting in the role of Cassandra Rushmore on the long-running Australian soap opera Neighbours in 1994. In 1996–1997, she starred as Catherine O'Brien on the same show. She starred as pilot Fry in the science fiction movie Pitch Black (2000), which was filmed in Australia while playing the character with an American accent. She also made two guest appearances on the long-running Australian police drama Blue Heelers, first in the drama's debut episode in 1994 and again in 1996.

Woody Allen personally called her mobile phone to offer her the role of Melinda in Melinda and Melinda (2004). She was convinced that it was a prank until the script was delivered to her. Mitchell was also a lead character in the movie Mozart and the Whale (2005), a story about two people with Asperger's Syndrome trying to make a go of their relationship despite their condition.
